Point mutation. Computer artwork of a single strand of DNA (deoxyribonucleic acid) with a point mutation (illuminated). A point mutation is caused by the replacement of a single base nucleotide (red, blue, green or yellow) with one of the three other nucleotides. The sequence of bases along the strands forms a template for the production of proteins. Therefore, the change in base in a point mutation can lead to a different amino acid being incorporated into the encoded protein, often significantly changing its properties. An example is the disease sickle-cell anaemia, which is caused by a point mutation in the gene coding for the blood protein beta-haemoglobin
